IL-1 cluster gene polymorphisms in Turkish patients with gout and their association with ınflammatory markers: a cross-sectional genetic study

Objective. Gout is an autoinflammatory disorder driven by monosodium urate crystal–induced activation of interleukin-1 (IL-1) signaling. Despite the central role of IL-1 in the inflammatory cascade, data on IL-1 gene polymorphisms in Turkish patients with gout are scarce. We aimed to evaluate variants within the IL-1 gene cluster and to assess their potential associations with inflammatory markers. Methods. Seventy gout patients and 106 age- and sex-matched healthy controls were included. Genomic deoxyribonucleic acid (DNA) extracted from peripheral blood was genotyped for five IL-1 single-nucleotide polymorphisms (rs16944, rs1143634, rs1800587, rs2234650, rs315952) using real-time polymerase chain reaction (PCR) with melting-curve analysis. Genotype and allele frequencies were compared between groups, and laboratory variables were assessed across genotypes within the patient cohort. Statistical significance was set at p < 0.05. Results. The rs16944 AG genotype was significantly more common in gout patients than in controls.
