Direct Fluorometric Determination of Liver Alcohol Dehydrogenase with Berberine

A fluorometric method for the determination of the concentration of horse liver alcohol dehydrogenase is described. It is based on the formation of a strongly fluorescent complex between the enzyme and berberine. The relative increase of fluorescence after the addition of the enzyme into a solution of berberine (at fixed concentration) is a linear function of the enzyme concentration.The results are easy to obtain, nearly temperature independent and as reliable as those of the commonly used activity measurement.The principle of this method can also be used for the estimation of other proteins forming strongly fluorescent complexes with slightly fluorescent probes.
