Enzymatic Catalysis in Solidified Media

Water insoluble blue starch polymer particles incorporated into agar medium were shown to be hydrolyzed by α‐amylase enzyme. When a drop of α‐amylase was placed on such a medium the enzyme diffused at the same time hydrolyzing the water insoluble blue starch polymer to form a clear area which was easily visible and easily measured. The size of the hydrolyzed area was studied with respect to the time of hydrolysis, temperature of hydrolysis, and enzyme concentration. The variance of repeated measurements of the diameter of 80 hydrolyzed areas, as well as one area measured ten times, was determined. The variance in diameter measurements of the hydrolyzed areas on different plates and at different distances from the center of the plate was determined.
