Construction of Isokinetic Sucrose Gradients for Rate‐Zonal Centrifugation

An isokinetic gradient is a density and viscosity gradient, in which the particles during the centrifugation sediment with constant velocity. A numerical method for construction of isokinetic sucrose gradients for gradient centrifugation in B XIV zonal rotors is described, and several examples of gradients for separation of soluble proteins, nucleic acids, ribosomes and polysomes are given. The general usefulness of isokinetic gradients is illustrated by experimental separations of soluble immune complexes and of rat brain RNA.
