AbstractProteins involved in signal transduction are often phosphorylated. Determination of the specific amino acid residue(s) involved is used in characterizing the particular pathway. Partial acid hydrolysis of phosphorylated proteins followed by two‐dimensional thin layer chromatography is used to identify the phosphorylated residues of the protein as phosphoserine, phosphothreonine, or phosphotyrosine. Mild alkaline hydrolysis is used to enhance detection of phosphothreonine and phosphotyrosine.
